Bu ba\u011flamda T\u00fcrkiye&#8217;de \u00dcsk\u00fcdar \u00dcniversitesinin Transgenik H\u00fccre Teknolojileri ve Epigenetik Uygulama ve Ara\u015ft\u0131rma Merkezi&#8217;nde de kanser ve nadir hastal\u0131klara y\u00f6nelik laboratuvar \u00e7al\u0131\u015fmalar\u0131 y\u00fcr\u00fct\u00fcl\u00fcyor.<\/p>\n<p>\u00dcsk\u00fcdar \u00dcniversitesi Rekt\u00f6r Yard\u0131mc\u0131s\u0131 ve Molek\u00fcler Biyoloji Ana Bilim Dal\u0131 Ba\u015fkan\u0131 Prof. Dr. Muhsin Konuk ile M\u00fchendislik ve Do\u011fa Bilimleri Fak\u00fcltesi Molek\u00fcler Biyoloji ve Genetik B\u00f6l\u00fcm\u00fc Dr. \u00d6\u011fretim \u00dcyesi Cihan Ta\u015ftan, AA muhabirinin, gen d\u00fczenleme y\u00f6ntemine ili\u015fkin sorular\u0131n\u0131 yan\u0131tlad\u0131.<\/p>\n<p>Prof. Dr. Konuk, &#8220;gen d\u00fczenleme&#8221;, &#8220;gen editleme&#8221; ve &#8220;gen cerrahisi&#8221; olarak isimlendirilen CRISPR&#8217;\u0131n genler i\u00e7erisinde bozuk olan b\u00f6lgenin d\u00fczeltilmesi i\u00e7in kullan\u0131ld\u0131\u011f\u0131n\u0131 belirtti.<\/p>\n<p><img decoding=\"async\" alt=\"\" src=\"https:\/\/www.aa.com.tr\/uploads\/userFiles\/a54ea27f-6036-4f79-88c6-6bc9ba19a35d\/08_Nisan%2F06%20temmuz%2F12%2FAA-31907992.jpg\"\/><\/p>\n<h3>Ba\u015far\u0131 oran\u0131 \u00e7ok y\u00fcksek ama &#8220;etik y\u00f6n\u00fc&#8221; tart\u0131\u015f\u0131l\u0131yor<\/h3>\n<p>CRISPR&#8217;dan \u00f6nce gen d\u00fczenleme teknolojisi olarak iki farkl\u0131 y\u00f6ntemin bulundu\u011funu ancak bunlar\u0131n \u00e7al\u0131\u015fma mekanizmas\u0131n\u0131n daha uzun s\u00fcrd\u00fc\u011f\u00fcn\u00fc ve hata verme oranlar\u0131n\u0131n y\u00fcksek \u00e7\u0131kt\u0131\u011f\u0131n\u0131 s\u00f6yleyen Konuk, bu teknikteki ba\u015far\u0131 oran\u0131n\u0131n ise y\u00fczde 99&#8217;un \u00fcst\u00fcnde oldu\u011funa i\u015faret etti.<\/p>\n<p>Prof. Dr. Muhsin Konuk, bu y\u00f6ntemin embriyonun geneti\u011finin de\u011fi\u015ftirilmesi i\u00e7in kullan\u0131lmas\u0131n\u0131n hem etik hem bilimsel a\u00e7\u0131dan ciddi problemler ortaya \u00e7\u0131karabilece\u011finin d\u00fc\u015f\u00fcn\u00fcld\u00fc\u011f\u00fcn\u00fc dile getirdi.<\/p>\n<p>CRISPR gen d\u00fczenleme y\u00f6nteminin baz\u0131 \u00fclkelerce etik kabul etmedi\u011fini aktaran Konuk, &#8220;Baz\u0131 \u00fclkeler \u00f6zellikle insan embriyosu ve insan \u00fcreme h\u00fccrelerinin CRISPR&#8217;la d\u00fczenlenmesi i\u00e7in sadece deneysel \u00e7al\u0131\u015fmalar\u0131n yap\u0131labilece\u011fini \u00f6ng\u00f6r\u00fcyor. Deneysel \u00e7al\u0131\u015fma bittikten sonra da o h\u00fccrelerin ve embriyolar\u0131n ortadan kald\u0131r\u0131lmas\u0131 zorunlulu\u011funu getiriyor. Hangi tipte, g\u00f6z ve sa\u00e7 renginde ya da zeka d\u00fczeyinde insanlar\u0131n ortaya \u00e7\u0131kaca\u011f\u0131yla ilgili tart\u0131\u015fmalar \u015fimdiden ba\u015flad\u0131. \u0130nsan eliyle gen frekans\u0131n\u0131 ve genetik havuzda olmas\u0131 gereken \u015feyleri de\u011fi\u015ftirme yoluna da girilebilir. Bu y\u00f6nleriyle etik a\u00e7\u0131dan tart\u0131\u015f\u0131l\u0131yor.&#8221; diye konu\u015ftu.<\/p>\n<h3>Genetikleri de\u011fi\u015ftirilmi\u015f bebekler bilim d\u00fcnyas\u0131n\u0131 aya\u011fa kald\u0131rd\u0131<\/h3>\n<p>Dr. \u00d6\u011fretim \u00dcyesi Cihan Ta\u015ftan ise CRISPR gen m\u00fchendisli\u011fiyle bir\u00e7ok canl\u0131n\u0131n geneti\u011finin de\u011fi\u015ftirilebilece\u011finin anla\u015f\u0131ld\u0131\u011f\u0131n\u0131 belirtti.<\/p>\n<p>Alt\u0131 y\u0131l \u00f6nce geneti\u011fi de\u011fi\u015ftirilen embriyolar\u0131n t\u00fcp bebek y\u00f6ntemiyle anne rahmine yerle\u015ftirilmesiyle iki bebe\u011fin d\u00fcnyaya geldi\u011fini hat\u0131rlatan Ta\u015ftan, bu iki bebe\u011fin HIV vir\u00fcs\u00fcn\u00fcn neden oldu\u011fu AIDS&#8217;e kar\u015f\u0131 diren\u00e7li olaca\u011f\u0131n\u0131n s\u00f6ylenildi\u011fini kaydetti.<\/p>\n<p>Bu konunun bilim d\u00fcnyas\u0131nda tepkiye yol a\u00e7t\u0131\u011f\u0131n\u0131 anlatan Ta\u015ftan, &#8220;Tabii bu HIV gibi bir hastal\u0131\u011f\u0131n tedavisi noktas\u0131nda d\u00fc\u015f\u00fcn\u00fclebilir. Ama hayat\u0131 tehlikeye atabilecek, o bebekler b\u00fcy\u00fcd\u00fckten sonra \u00f6lmelerine sebep olabilecek bir\u00e7ok genetik hastal\u0131\u011f\u0131n da embriyo d\u00fczeyindeyken CRISPR&#8217;la tedavi edilebilece\u011fine dair ipucunu da bize sundu.&#8221; de\u011ferlendirmesini yapt\u0131.<\/p>\n<p>Ta\u015ftan, hen\u00fcz embriyonik d\u00fczenlemenin d\u00fcnyan\u0131n her yerinde yasak oldu\u011funu vurgulayarak, bir insan\u0131n do\u011fduktan sonra kalp rahats\u0131zl\u0131klar\u0131na yakalan\u0131p hayat\u0131n\u0131 kaybetmesine sebep olabilecek genetik DNA fakt\u00f6rlerinin CRISPR y\u00f6ntemiyle embriyo d\u00fczeyinde tedavi edilebilecek olmas\u0131n\u0131n ise umut verici oldu\u011fu yorumunda bulundu.<\/p>\n<h3>&#8220;Kanseri tan\u0131y\u0131p \u00f6ld\u00fcrebilen CRISPR ila\u00e7lar\u0131 geli\u015ftirilebiliyor&#8221;<\/h3>\n<p>Dr. \u00d6\u011fr. \u00dcyesi Cihan Ta\u015ftan, gen d\u00fczenlemenin hangi hastal\u0131klar\u0131n tedavisinde kullan\u0131labilece\u011fine y\u00f6nelik \u015funlar\u0131 kaydetti:<\/p>\n<p>&#8220;\u0130nsan v\u00fccudunda 75 bin farkl\u0131 genetik bozuklu\u011fun hastal\u0131klara sebep oldu\u011fu d\u00fc\u015f\u00fcn\u00fcl\u00fcyor. Bu bozukluklar d\u00fcnyada nadir ya da genetik hastal\u0131k olarak adland\u0131r\u0131lan 7 bin farkl\u0131 rahats\u0131zl\u0131\u011f\u0131n ortaya \u00e7\u0131kt\u0131\u011f\u0131n\u0131 g\u00f6sterdi. Son 5 y\u0131lda \u00f6zellikle CRISPR gen m\u00fchendisli\u011findeki geli\u015fmeler bize, bu 7 bin hastal\u0131\u011f\u0131n yakla\u015f\u0131k y\u00fczde 89&#8217;unun tedavi edilebilece\u011fini g\u00f6sterdi. Bu durum SMA ve DMD&#8217;den tutun bir\u00e7ok genetik kaynakl\u0131 hastal\u0131\u011f\u0131n art\u0131k bu m\u00fchendislikle tedavi edilebilece\u011fini g\u00f6steriyor. \u015eu an d\u00fcnyada 100&#8217;den fazla insan CRISPR&#8217;la geneti\u011fi de\u011fi\u015ftirilmi\u015f tedaviler al\u0131yor, yani klinik denemeler de ba\u015flam\u0131\u015f durumda.&#8221;<\/p>\n<p>Bir insan d\u00fcnyaya geldikten sonra gen d\u00fczenlemesinin m\u00fcmk\u00fcn olup olmad\u0131\u011f\u0131na ili\u015fkin Ta\u015ftan, \u015f\u00f6yle konu\u015ftu:<\/p>\n<p>&#8220;Baz\u0131 genetik hastal\u0131klar var ki ki\u015fi do\u011fduktan sonra da bunlar\u0131 tamir edebiliyoruz. Bunlar\u0131n en bilinenlerinden k\u00f6k h\u00fccreleri tamir edebildi\u011fimizde kan hastal\u0131klar\u0131, n\u00f6romusk\u00fcler hastal\u0131klar, SMA ve DMD gibi hastal\u0131klar\u0131n temelinde yatan h\u00fccreleri nanorobotlarla, CRISPR&#8217;\u0131 ta\u015f\u0131yan partik\u00fcllerle, nano lipidlerle hedefleyebiliyoruz ve o h\u00fccrelerin geneti\u011fini v\u00fccut i\u00e7erisindeyken de\u011fi\u015ftirebiliyoruz. Bununla beraber kanseri \u00f6ld\u00fcrme noktas\u0131nda kanseri hedefleyebiliyoruz. Klasik kimyasal ila\u00e7lar yerine art\u0131k sadece kanseri tan\u0131y\u0131p \u00f6ld\u00fcrebilen CRISPR ila\u00e7lar\u0131 geli\u015ftirilebiliyor. \u00c7oklu antibiyotik diren\u00e7li bakteriler de var ve en \u00f6nemli \u00f6l\u00fcmc\u00fcl sebeplerden biri.&#8221;<\/p>\n<h3>\u015eizofreni sendromlar\u0131 azalt\u0131labilir<\/h3>\n<p>Dr. \u00d6\u011fr. \u00dcyesi Ta\u015ftan, \u015fu ana kadarki \u00e7al\u0131\u015fmalarda, CRISPR y\u00f6nteminin bu hastal\u0131klar\u0131n yakla\u015f\u0131k y\u00fczde 90&#8217;\u0131n\u0131 tamir edebilece\u011fini g\u00f6sterdi\u011fini ancak hala 1000 hastal\u0131\u011f\u0131n tedavisinin m\u00fcmk\u00fcn olmad\u0131\u011f\u0131n\u0131 \u00e7\u00fcnk\u00fc b\u00fcy\u00fck genetik elementler ya da DNA fakt\u00f6rlerinin bir insanda silinmi\u015f olmas\u0131n\u0131n \u015fu anki teknolojilerle de d\u00fczeltilemeyecek bir durum oldu\u011funu anlatt\u0131.<\/p>\n<p>Beyine de m\u00fcdahale edilebildi\u011fine dikkati \u00e7eken Ta\u015ftan, &#8220;\u015eu an \u015fizofreni ile parkinson, Alzheimer gibi n\u00f6rolojik hastal\u0131klarda da CRISPR veya gen tedavilerinin yap\u0131labilece\u011fiyle ilgili bir\u00e7ok \u00e7al\u0131\u015fma bulunuyor. \u015eizofreni ve otizm gibi hastal\u0131klar hem DNA&#8217;lar\u0131ndan etkilenebilen hem de DNA&#8217;dan ba\u011f\u0131ms\u0131z fakt\u00f6rlerin oldu\u011fu hastal\u0131klar. Ama son \u00e7al\u0131\u015fmalar \u015fizofreni hastalar\u0131n\u0131n da baz\u0131 genetik fakt\u00f6rlerini tamir edebildi\u011fimizde sendromlar\u0131n\u0131 azaltabilece\u011fimizi \u00f6ng\u00f6r\u00fcyor.&#8221; ifadelerini kulland\u0131.<\/p>\n<p>Ta\u015ftan, T\u00fcrkiye&#8217;deki bir\u00e7ok farkl\u0131 laboratuvarda CRISPR&#8217;\u0131n insan hastal\u0131klar\u0131, tar\u0131m teknolojileri ve hayvanlarda da kullan\u0131labilece\u011fine dair \u00e7al\u0131\u015fmalar\u0131n s\u00fcrd\u00fcr\u00fcld\u00fc\u011f\u00fcnden bahsetti.
